Gabapentin
Gabapentin is a foundational structural analog of the neurotransmitter GABA, engineered to cross the blood-brain barrier with high efficiency. Like pregabalin, its primary mechanism of action relies on binding with high affinity to the regulatory \(\alpha_2\delta-1\) subunits of presynaptic voltage-dependent calcium channels. This binding attenuates calcium-mediated vesicular neurotransmitter exocytosis, soothing hyperexcited nociceptive signals and central seizure foci without interacting directly with GABA-A or GABA-B receptors.
