Glibenclamide
Glibenclamide is a classic, highly potent second-generation sulfonylurea hypoglycemic drug. It targets and binds tightly to the sulfonylurea receptor 1 (SUR1) subunits of ATP-sensitive potassium channels on pancreatic beta-cells. The resulting channel closure causes structural cell depolarization, opening voltage-gated calcium channels to force a strong, sustained release of pre-formed insulin, effectively controlling both fasting and post-meal plasma glucose concentrations.
